Spotify offers a free, ad-supported unlimited account and an ad-free premium account. The unlimited account includes intermittent audio ads and graphical ads embedded in the application. The only way to remove Spotify ads is to upgrade your free account to a premium subscription, which costs $9.99 per month as of January 2014.
Mute Spotify Audio Ads
While you can't avoid overlay ads, billboard ads or homepage takeovers with a free account, you can install software that automatically mutes audio ads. Blockify for Windows is a simple application that mutes Spotify when it detects an ad and plays an audio file from your computer during the interval (link in Resources). Smutefy and SpotiFree for Mac OS X mute Spotify during ads but don't substitute audio from your computer (links in Resources). Setup for each of these applications is simple, as you need only to run the program while playing a Spotify playlist. The software automatically detects the open Spotify application and mutes it during ad breaks.
